ABSTRACT

Objective:To investigate the value of combined detection of serum neurogranin (NG) and hypoxia-inducible factor-1α (HIF-1α) in patients with severe craniocerebral trauma.Methods:Ninety-seven patients with severe craniocerebral trauma from June 2018 to March 2020 in Jinshan Branch of Shanghai Sixth People′s Hospital were selected. According to the Glasgow outcome score (GOS), 97 patients were divided into good prognosis group (GOS>3 scores, 46 cases) and poor prognosis group (GOS ≤ 3 scores, 51 cases). The NG, HIF-1α, Glasgow coma score (GCS), acute physiology and chronic health status score Ⅱ (APACHE Ⅱ) were compared between 2 groups. The independent risk factors of prognosis in patients with severe craniocerebral trauma were analyzed by multivariate Logistic regression analysis. The diagnostic efficacy of NG and HIF-1α on poor prognosis in patients with severe craniocerebral trauma was analyzed by receiver operating characteristic (ROC) curve. The correlation between serum NG, HIF-1α and APACHE Ⅱ in patients with severe craniocerebral trauma was analyzed by Pearson analysis.Results:The GCS in good prognosis group was significantly higher than that in poor prognosis group: (6.50 ± 1.74) scores vs. (4.76 ± 0.78) scores, the NG, HIF-1α and APACHE Ⅱwere significantly lower than those in poor prognosis group: (696.98 ± 158.96) ng/L vs. (875.92 ± 188.52) ng/L, (34.72 ± 13.98) μg/L vs. (51.29 ± 14.17) μg/L and (15.69 ± 3.45) scores vs. (22.58 ± 6.45) scores, and there were statistical differences ( P<0.01). Multivariate Logistic regression analysis result showed that the NG, HIF-1α, APACHEⅡ, GCS and type of craniocerebral trauma were independent risk factors on the prognosis in patients with severe craniocerebral trauma ( P<0.05 or<0.01). ROC curve analysis result showed that the AUC of NG and HIF-1αNG and HIF-1α combined detection to assess the poor prognosis in patients with severe craniocerebral trauma was significantly higher than NG and HIF-1α alone detection (0.873 vs. 0.772 and 0.821, Z = 2.276 and 1.949, P<0.05). Pearson correlation analysis result showed that APACHE Ⅱ was positive correlation with serum NG and HIF-1α in severe craniocerebral trauma patients with poor or good prognosis ( r = 0.852 and 0.889, P<0.01; r = 0.717 and 0.851, P<0.01). Conclusions:The combined detection of serum NG and HIF-1α can be used as an evaluation index for the prognosis in patients with severe craniocerebral trauma, which helps to determine the severity of craniocerebral trauma and has great value for clinical diagnosis and treatment.

ABSTRACT

Neurogranin (Ng), as a neuron-specific postsynaptic protein, is abundant in the cerebral cortex and hippocampus. Ng may alter the affinity with calmodulin through phosphorylation and redox, and participate in the calcium signal pathway through glutamate receptor, to regulate the information transfer and synaptic plasticity. Ng involves in the formation and encoding of memories in related brain regions and plays an important role in learning, memory and cognitive function. Ng is associated with cognitive decline in Alzheimer's disease and can be used as a synaptic biomarker for diagnosis. Cognitive dysfunction after stroke is associated with decrease of Ng expression in the brain. Ng is one of the sites of genetic variants associated with susceptibility to schizophrenia.

ABSTRACT

@#Objective To study the effect of exploratory environment on behavioral recovery and the expression of neurogranin (Ng) in rats after unilateral local cerebral infarction. Methods 70 male Sprague-Dawley rats were adopted. After making the model of MCAO with electric coagulation, 60 rats were randomly divided into exploratory group (n=30, living in maze cages) and control group (n=30, living in large standard cages). Other 10 rats as the sham group lived in standard cage. 10 rats were chosen randomly from the groups for Berderson test, manual muscle and balancing test on the 1st, 7th, 14th, 28th day after operation, and 5 rats were sacrificed separately in exploratory group and control group at the same time. 5 rats in sham group were randomly sacrificed separately on the 7th, 28th day after operation. The expressions of Ng in the peri-ischemic cortex were detected with immunohistochemistry stain. Results The scores of behavior in exploratory group improved compared with that in control group (P<0.05) from 14 d after operation. The number of Ng positive cells increased in exploratory group compared with that in control group (P<0.01) from 7 d after operation. Conclusion Exploratory environment can facilitate the functional recovery and the Ng expression in rats after unilateral local cerebral infarction.

ABSTRACT

Objective To investigate the mRNA expression level of neurogranin on peripheral blood mononuclear cells(PBMCs)from patients with systemic lupus erythematosus(SLE).Methods Top 20 tags of SLE PBMCs SAGE library were searched from normal lymphocytes SAGE library including navie-T,Th1,Th2, CD8+T, NK and B cells,and their abundance was compared.The mRNA expression level of neuro-granin,a differential over-expressed tag,was detected in 35 cases of SLE and 15 normal controls by reversetranscription-polymerase chain reaction (RT-PCR).Results Neurogranin tag could only be detected in SLE PBMCs SAGE library,but was hardly found in normal lymphocyte SAGE library.However,either SLE pa-tients or normal controls showed a detectable mRNA level of neurogranin on PBMCs by RT-PCR.The mRNA level of neurogranin in active SLE patients was significantly increased than those in controls(P<0.001).but only slightly increased in inactive SLE patients (P>0.05).Conclusion Neurogranin,as a novel proapototic factor,is overexpressed on PBMCs of SLE patients.It may be involved in the regulation of abnormal immune responses in lupus.

ABSTRACT

Adult male Kunming mice were divided into normal control group,propylthiouracil(PTU) treated group and PTU+T_4 treated group.Neurogranin(Ng)protein expression in the hippocampus after 8 weeks was assayed by Western blotting and immunohistochemical method.Results showed that Ng protein was decreased in the hippocampus of male mice with adult-onset hypothyroidism,suggesting that decreased Ng,which is correctable by T_4,is involved in cognitive dysfunction in adult-onset hypothyroid male mice.

AIM:To study the effects of different duration of sleep deprivation (SD) on neurogranin (Ng),protein kinase C (PKC) and Ca2+-calmodulin dependent protein kinase II (CaMK II) mRNA expressions in hippocampus and forebrain of rats. METHODS:Male Wistar rats were divided into three groups:24 h,48 h and 72 h experimental groups. Each group was divided into sleep deprivation group (SD group) and control group (C group). SD model was established by sleep deprivation box. One step methods by Trizol was used to extract hippocampus and forebrain neuronal total RNA. ABSTRACT
